Amphiphilic Macromolecules on Cell Membranes: From Protective Layers to Controlled Permeabilization.
International audienceAntimicrobial and cell-penetrating peptides have inspired developments of abiotic membrane-active polymers that can coat, penetrate, or break lipid bilayers in model systems. Application to cell cultures is more recent, but remarkable bioactivities are already reported. Synthetic polymer chains were tailored to achieve (i) high biocide efficiencies, and selectivity for bacteria (Gram-positive/Gram-negative or bacterial/mammalian membranes), (ii) stable and mild encapsulation of viable isolated cells to escape immune systems, (iii) pH-, temperature-, or light-triggered interaction with cells. This review illustrates these recent achievements highlighting the use of abiotic polymers, and compares the major structural determinants that control efficiency of polymers and peptides. Charge density, sp. of cationic and guanidinium side groups, and hydrophobicity (including polarity of stimuli-responsive moieties) guide the design of new copolymers for the handling of cell membranes. While polycationic chains are generally used as biocidal or hemolytic agents, anionic amphiphilic polymers, including Amphipols, are particularly prone to mild permeabilization and/or intracell delivery

Induction of Paralysis and Visual System Injury in Mice by T Cells Specific for Neuromyelitis Optica Autoantigen Aquaporin-4.
While it is recognized that aquaporin-4 (AQP4)-specific T cells and antibodies participate in the pathogenesis of neuromyelitis optica (NMO), a human central nervous system (CNS) autoimmune demyelinating disease, creation of an AQP4-targeted model with both clinical and histologic manifestations of CNS autoimmunity has proven challenging. Immunization of wild-type (WT) mice with AQP4 peptides elicited T cell proliferation, although those T cells could not transfer disease to naïve recipient mice. Recently, two novel AQP4 T cell epitopes, peptide (p) 135-153 and p201-220, were identified when studying immune responses to AQP4 in AQP4-deficient (AQP4-/-) mice, suggesting T cell reactivity to these epitopes is normally controlled by thymic negative selection. AQP4-/- Th17 polarized T cells primed to either p135-153 or p201-220 induced paralysis in recipient WT mice, that was associated with predominantly leptomeningeal inflammation of the spinal cord and optic nerves. Inflammation surrounding optic nerves and involvement of the inner retinal layers (IRL) were manifested by changes in serial optical coherence tomography (OCT). Here, we illustrate the approaches used to create this new in vivo model of AQP4-targeted CNS autoimmunity (ATCA), which can now be employed to study mechanisms that permit development of pathogenic AQP4-specific T cells and how they may cooperate with B cells in NMO pathogenesis
Measurement and Compensation of Horizontal Crabbing at the Cornell Electron Storage Ring Test Accelerator
In storage rings, horizontal dispersion in the rf cavities introduces
horizontal-longitudinal (xz) coupling, contributing to beam tilt in the xz
plane. This coupling can be characterized by a "crabbing" dispersion term
{\zeta}a that appears in the normal mode decomposition of the 1-turn transfer
matrix. {\zeta}a is proportional to the rf cavity voltage and the horizontal
dispersion in the cavity. We report experiments at the Cornell Electron Storage
Ring Test Accelerator (CesrTA) where xz coupling was explored using three
lattices with distinct crabbing properties. We characterize the xz coupling for
each case by measuring the horizontal projection of the beam with a beam size
monitor. The three lattice configurations correspond to a) 16 mrad xz tilt at
the beam size monitor source point, b) compensation of the {\zeta}a introduced
by one of two pairs of RF cavities with the second, and c) zero dispersion in
RF cavities, eliminating {\zeta}a entirely. Additionally, intrabeam scattering
(IBS) is evident in our measurements of beam size vs. rf voltage.Comment: 5 figures, 10 page
A test for the search for life on extrasolar planets: Looking for the terrestrial vegetation signature in the Earthshine spectrum
We report spectroscopic observations (400 to 800nm, R = approx 100) of
Earthshine in June, July and October 2001 from which normalised Earth albedo
spectra have been derived. The resulting spectra clearly show the blue colour
of the Earth due to Rayleigh diffusion in its atmosphere. They also show the
signatures of oxygen, ozone and water vapour. We tried to extract from these
spectra the signature of Earth vegetation. A variable signal (4 to 10 +/-3%)
around 700nm has been measured in the Earth albedo. It is interpreted as being
due to the vegetation red edge, expected to be between 2 to 10% of the Earth
albedo at 700nm, depending on models. We discuss the primary goal of the
present observations: their application to the detection of vegetation-like
